| Morphological Characteristics | The colonies are circular with entire margins, opaque, and convex. The surface is smooth, shiny, moist, and easily lifted. No pigment is produced. The obverse side is white (light-colored). Microscopic examination reveals Gram-positive (staining blue-violet) cocci. The culture is pure. |
| Culture Method | ① Prepare one test tube containing 5-10 mL of liquid medium and two agar plates.② Within a biological safety cabinet, sterilize and open the ampoule vial. Briefly flame the top, then immediately apply a drop of sterile water to crack it. Use sterile forceps to break it open.③ Aspirate 0.5 mL of the liquid medium and inject it into the freeze-dried vial. After thorough resuspension, transfer the entire volume back into the liquid medium test tube and mix well.④ Aspirate 0.2 mL of the bacterial suspension and inoculate it onto an agar plate. Spread evenly. Repeat this step to prepare a second Agar plate.⑤ Incubate both the liquid culture tube and all agar plates under the aforementioned conditions. The culture is ready for use once growth is observed. |
