Culture MethodUse a Pasteur or 1.0 mL pipette to extract approximately 0.5 to 1.0 mL from a tube containing 2.5 mL of # 1092 broth. Re hydrate the entire particle. Aseptically transfer these components back into the tube. Stir to make it mixed evenly. Dilute continuously by transferring 0.25 milliliters of liquid from the original tube to a tube containing 2.25 milliliters. Repeat this process, transfer 0.25 milliliters from the second tube to the third tube, and so on. Dilution is important not only for titration, but also for maintaining the culture at different growth stages. Many strains of bacteria will quickly die once they reach acidic or alkaline conditions. It is recommended to prepare multiple diluents from the initial tube, as the low-temperature protectants used during freeze-drying often inhibit growth. Perform aerobic incubation of all tubes and purity plates at 37 ° C. Depending on the medium used, growth may manifest as increased turbidity, color change, or a combination of both.
