| Culture Method | ① Inoculation procedure: 24-48 hours prior to inoculation, seed host cells in T25 flasks. Prepare for viral inoculation when host cell density reaches 80-90%; Remove the cell culture medium and inoculate 100-500μL of virus solution. Allow adsorption for 1-2 hours under culture conditions, gently shaking the T25 flask every 15-30 minutes to ensure even virus distribution. Add 6mL of virus growth medium to complete adsorption, then transfer to the incubator for continued culture. Regularly observe for cytopathic effects; ② Virus Harvesting: Initiate virus collection once host cells exhibit complete cytopathic changes. During collection, freeze-thaw the cells 1–2 times, then filter through a 0.22μm pore size filter. The collected filtrate constitutes the virus solution. |
