Culture Method(1)Prepare 1–2 agar plate medium as described above. (2) Open the ampoule in the biosafety cabinet, heat the tip of ampoule in a flame, quickly drop sterile water to creak it, then break it with forceps; (3)Add 0.5 mL sterile water to the freeze dried ampoule, mix thoroughly to reconstitute the lyophilized pellet, then pipette 200 μL onto each plate and spread evenly. (4) Incubate plates under the specified growth conditions; the strain is ready for use once growth is observed.
