Culture MethodRecovery steps: ① Remove the cryovial from liquid nitrogen or -80 °C freezer and place it in PE gloves. Quickly immerse it in a 37 °C water bath and shake the cryovial to accelerate dissolution. It is recommended to dissolve it completely within 1 minute; ② Add the dissolved cell solution into a centrifuge tube containing 9mL of complete culture medium on a super clean bench, centrifuge at 1000-1200rpm for 5 minutes, discard the supernatant, and resuspend the cells in 1-2mL of complete culture medium. ③ Then add the cell suspension to a T25 bottle containing 6-8mL of complete culture medium and culture it in an incubator.Cell passage: ① Gently scrape off the adherent cells with a cell scraper, transfer them along with the culture medium to a centrifuge tube, centrifuge at 1000 RPM for 5-8 minutes, and discard the supernatant; ② Add 1-2 mL of complete culture medium to the cell pellet collected from suspended cells and adherent cells, and mix well; ③ Add culture medium at a rate of 5-6mL/bottle, and divide the cell suspension into culture bottles containing 5-6mL of culture medium at a ratio of 1:2; ④ Pay attention to changes in the pH value of the culture medium and cell density, change the medium regularly (2-3 times a week), and repeat the passage operation or cryopreservation when the cell density reaches 80% -90%.
