| Culture Method | ① Prepare 4-5 test tubes containing 4.5mL of prepared mycoplasma culture medium (with added serum and penicillin); ② Open the safety cabinet, burn the top with an alcohol lamp, quickly drip sterile water to break it, and then use tweezers to crush it; ③ Transfer 0.5mL of liquid culture medium into a freeze-drying tube, dissolve it thoroughly, and then transfer it back into the first liquid test tube; ④ Take 0.5mL of suspension from the first test tube and transfer it to the second test tube; ⑤ Repeat the above steps and dilute the gradient to the last tube (note: one tube needs to be left as a blank control). ⑥ Cultivate under the above culture conditions, and the strain can be used once it grows (the color of the culture tube changes from orange to red). |
