Culture Method① Prepare 1-2 plates as described above; ② Disinfect ampoule surface, open in safety cabinet, cauterize top with alcohol lamp, then promptly add sterile water to rupture. Crush with forceps; ③ Pipette 0.5mL sterile water into lyophilized tube. After complete dissolution, dispense 200μL/well onto plates and spread evenly; ④ Incubate plates under specified conditions until colonies emerge.
