| Culture Method | ① Prepare 1–2 plates as described above; ② Open in a safety cabinet, scorch the top with a Bunsen burner, then quickly add a drop of sterile water to rupture it, and subsequently crush it with forceps; ③ Pipette 0.5 mL of sterile water into a freeze-dried vial, dissolve the bacterial powder thoroughly, then dispense approximately 200 μL onto each of the two plates, spreading evenly; ④ Incubate the plates under the specified conditions; the culture is ready for use once growth appears. |
