Culture Method① Prepare 1-2 plates as described above; ② Disinfect the surface of test tube slants before opening in a biosafety cabinet; ③ Cut blocks (see appendix) into 0.5×0.5 cm² squares using sterile inoculating forks and spatulas; ④ Place small square blocks flat onto the center of plates, on the agar surface; ⑤ Incubate plates upright under the specified conditions until growth appears.
