Culture Method① Prepare 1-2 composite PDA plates; ② Open test tubes on a Agar slant surface after disinfection in a safety cabinet; ③ Cut into 0.5×0.5cm small square blocks using sterile inoculating forks and spatulas (see appendix);2 cm² squares; ④ Place the small squares flat in the center of the Agar plate, on the agar surface; ⑤ Incubate the plates upright under the specified conditions; the culture is ready for use once growth appears.
