Culture MethodHost culture: 37 °C, aerobic, nutrient agar medium, 18-24h. Nutrient agar medium: 3.0g beef extract, 10.0g peptone, 5.0g NaCl, 20.0g agar (excluding liquid medium), 1.0L distilled water, pH 7.0. Sterilize at 121 °C for 15 minutes. Phage culture: ① Host bacterial preparation: Select a single colony of host bacteria from a Agar plate, dissolve it in 40-50 mL nutrient broth medium, shake at 37 °C, 160-180 rpm until OD600 is 0.1-0.4; ② Transfer: Transfer an appropriate amount of host bacterial solution into the Agar plate, wait for the host bacterial solution to dry, and then apply bacteriophages onto the Agar plate; ③ Cultivation: Place the petri dish in a 37 °C incubator overnight for cultivation; ④ Bacterial culture: After 24 hours, plaques grow on the agar plate. Wash the agar plate and bacteriophages completely into a 40mL triangular flask and incubate overnight at 37 °C and 160-180rpm on a shaker. ⑤ Filtration: After 16-18 hours, centrifuge the bacteriophage culture at 4000g for 10 minutes and filter the lysate using a 0.22 μ m sterile filter. The filtrate can be stored at 4 °C
