Culture Method① Prepare 1 vial of sterile water and 2 x 90mm resistant plates; ② Disinfect ampoule surface, open in biosafety cabinet, cauterize top with alcohol lamp, then immediately add sterile water to rupture; break ampoule with forceps; ③ Pipette 0.5 mL sterile water into lyophilized tube, dissolve completely, then Agar plate onto antibiotic-resistant plates (200 μL/Agar plate); ④ Incubate the resistance plates under specified conditions for 18-24 hours before observation (do not seal or wrap plates); ⑤ Streak plates and pick single colonies onto 50mL LB resistance liquid medium; incubate overnight at 37°C on a shaking incubator; ⑥ Extract plasmids from an appropriate volume of bacterial culture as required by the experiment
