| Culture Method | ① Prepare 1-2 plates as described above; ② Disinfect the ampoule surface, open it in a safety cabinet, cauterize the top with an alcohol lamp, then quickly add sterile water to rupture it. Immediately crush it with forceps; ③ Pipette 0.5 mL sterile water into the lyophilized tube. After fully dissolving the bacterial powder, dispense 200 μL/well onto plates and spread evenly; ④ Incubate plates under specified conditions until colonies emerge. |
