Morphological CharacteristicsColonies are 1–2 mm in diameter, circular, with smooth, well-defined edges; they are opaque, white on the upper surface, convex in the center, with a smooth, lustrous surface; they have a moist texture, are easily lifted, are G+ (blue-violet), oval in shape, and have a purity rating of “pure.”
Culture Method① Prepare 1–2 of the above agar plates (place in an anaerobic environment 24 hours in advance to remove oxygen); ② Open in a safety cabinet, scorch the top with a酒精 lamp, then quickly add a drop of sterile water to crack the surface, and subsequently crush it with forceps; ③ Pipette 0.5 mL of sterile water (previously placed in an anaerobic environment for 24 hours to remove oxygen) into the freeze-dried vial; after thoroughly dissolving the bacterial powder, dispense approximately 200 μL onto each of the two plates and spread evenly; ④ Incubate the plates under the aforementioned conditions; the culture is ready for use once colonies appear.
