| Culture Method | ① Prepare 2 plates as described above (place them in an anaerobic environment for 24 hours for equilibration); ② Disinfect the surface of the ampoule tube and open it in a safety cabinet. Burn the top with an alcohol lamp, then quickly apply sterile water to crack it, followed by breaking it with tweezers; ③ Aspirate approximately 0.5 mL of liquid medium or sterile water (placed in an anaerobic environment for 24 hours for equilibration) into the freeze-dried tube, thoroughly dissolve the bacterial powder, and then inoculate onto 2 plates, 200 µL per Agar plate, spreading evenly; ④ Incubate the plates under the aforementioned culture conditions. |
