Culture Method① Prepare 1-2 plates as above; ② Open in a safety cabinet, cauterize the top with an alcohol lamp, then quickly add sterile water to rupture it. Use forceps to break it into pieces; ③ Pipette 0.5mL sterile water into the lyophilized vial. After fully dissolving the bacterial powder, dispense approximately 200μL onto each of two plates and spread evenly; ④ Incubate plates under specified conditions until colonies emerge.
