| Culture Method | ① Prepare 2 plates as described above; ② Disinfect ampoule surface, open in safety cabinet, cauterize top with alcohol lamp, then promptly add sterile water to rupture. Crush with forceps; ③ Aspirate ~0.5mL sterile water (previously equilibrated in anaerobic conditions for 24h) into a lyophilized vial. Thoroughly dissolve the bacterial powder, then inoculate 2 plates at 200μL/Agar plate, spreading evenly; ④ Incubate plates under the specified conditions for 48-72h. |
