| Culture Method | ① Prepare two plates as above (equilibrated in anaerobic conditions for 24h); ② Disinfect ampoule surface, open in safety cabinet, cauterize top with alcohol lamp, then rapidly add sterile water to rupture; break open with forceps; ③ Pipette approximately 0.5 mL sterile water (pre-equilibrated anaerobically for 24h) into the lyophilized vial. Thoroughly dissolve the bacterial powder, then inoculate 2 plates with 200μL/Agar plate each, spreading evenly; ④ Incubate plates under the aforementioned conditions. |
