| Culture Method | ①Inoculation procedure: 24-48 hours before inoculation, transfer the frozen host cells into T25 flask, remove the cell culture medium when the host cell density reaches 80-90%, and inoculate 100-500 μL of virus solution. Adsorb under growth conditions for 1-2 hours, gently shake the T25 flask every 15-30 minutes to make the virus distribution more uniform; Add 6mL of virus growth medium to end adsorption, transfer to the incubator for further cultivation, and regularly observe the cytopathy; ② Virus collection: Once all host cells have developed lesions, virus liquid collection can begin. When collecting, the cells are repeatedly frozen and thawed 1-2 times, and then filtered through a 0.22 μ m pore size filter. The collected filtrate is the virus solution. |
