| Morphological Characteristics | The colonies are 1-2 mm in diameter, circular with entire margins, and opaque. The colony surface is cream-white, convex, smooth, shiny, and moist in texture. They are easily picked up. The cells are Gram-positive (blue-violet) and rod-shaped (bacilli). The culture is pure. |
| Culture Method | ① Prepare two of the aforementioned agar plates (pre‑equilibrated under anaerobic conditions for 24 hours).② After surface disinfection, open the ampoule in a biosafety cabinet. Flame the top of the ampoule with an alcohol burner, then quickly apply a drop of sterile water to crack it, and gently break the ampoule using sterile forceps.③ Aspirate 0.5 mL of sterile water (pre‑equilibrated under anaerobic conditions for 24 hours) and inject it into the lyophilized vial. Thoroughly resuspend the bacterial pellet, then inoculate 200 µL onto each of the two plates and spread evenly.④ Incubate the plates under the specified culture conditions. The culture is ready for use once growth is observed. |
