Morphological CharacteristicsThe colonies are 1-2 mm in diameter, circular with irregular margins, and opaque. The obverse side is creamy white, convex (raised at the center), with a smooth and moist texture. The cells are Gram-positive (staining blue-violet), rod-shaped. Purity: Pure.
Culture Method① Prepare 1-2 fresh agar plates as specified above (pre-reduced in an anaerobic environment for 24 hours).② Within a biological safety cabinet, surface-sterilize and open the glass ampoule. Briefly flame the neck, then immediately apply a drop of sterile water to induce thermal cracking. Carefully open it using sterile forceps.③ Aspirate 0.5 mL of sterile, pre-deoxygenated water (similarly pre-reduced for 24 hours) and inject it into the freeze-dried vial. Mix thoroughly for complete resuspension. Transfer the entire suspension into a tube containing liquid medium and mix well.④ Inoculate 0.2 mL of the resulting bacterial suspension onto each pre-reduced agar plate and spread evenly. Prepare a duplicate Agar plate following the same method.⑤ Incubate all plates under the aforementioned anaerobic conditions. The culture is ready for use when visible growth appears.
